The changing therapeutic landscape of small‑cell lung cancer
We summarise current standards of care, recent advances, and highlight emerging approaches showing early promise for better management of SCLC.
Small‑cell lung cancer (SCLC) is an exceptionally aggressive malignancy that is often metastatic at diagnosis. Although initially responsive to chemotherapy, responses are typically transient and recurrent disease has become largely refractory to standard cytotoxic agents. The past decade has brought major advances in our understanding of SCLC biology, and this preclinical progress is now informing multiple novel therapeutic approaches. Targeted therapies exploiting cell‑surface proteins uniquely or differentially expressed in SCLC—including T‑cell engagers, antibody‑drug conjugates, radioconjugates, and cell therapies—are showing substantial preliminary evidence of efficacy. In this series paper we summarize current standards of care, recent advances, and highlight emerging approaches showing early promise for better management of SCLC, providing new hope for patients with this particularly lethal disease.
SCLC remains one of the most lethal lung cancers, and identifying emerging therapies that can improve outcomes is critical for patients who have limited treatment options beyond initial chemotherapy.
