A study reported that small cell lung cancer (SCLC) cells depend on the nonsense‑mediated mRNA decay (NMD) pathway to tolerate their high mutation burden and evade immune detection. Pharmacological inhibition of the NMD kinase SMG1 induces accumulation of mutant proteins, causing tumor‑cell death and enhancing presentation of abnormal peptides on the cell surface, thereby making the tumor recognizable by cytotoxic T cells.

Targeting the NMD pathway, particularly the SMG1 kinase, can transform an immunologically “cold” SCLC tumor into one that is susceptible to T‑cell‑mediated immune attack, opening the door for new immunotherapy combinations and potentially improving outcomes for patients with this aggressive cancer.
