2025 World Conference on Lung Cancer (WCLC): key news stories
A first‑in‑human Phase 1 study of the DLL3‑directed antibody‑drug conjugate SHR‑4849 showed early signs of anti‑tumor activity in patients with relapsed small cell lung cancer.
The DeLLphi‑305 Phase III trial evaluated durvalumab (Imfinzi) plus tarlatamab versus durvalumab alone as first‑line maintenance therapy in patients with extensive‑stage small cell lung cancer (ES‑SCLC) who had not progressed after induction with durvalumab plus platinum/etoposide. Results showed a statistically significant and clinically meaningful improvement in overall survival (OS) with the combination, as well as in progression‑free survival (PFS) and objective response rate (ORR).
The study demonstrates that targeting DLL3, a protein over‑expressed in small cell lung cancer, can achieve meaningful objective responses and disease control, offering a new therapeutic option for patients with relapsed SCLC who have limited treatment choices.
