Mount Sinai Study Finds Chemotherapy-Free Treatment Shows Promise for Some Patients With Metastatic Breast Cancer
A chemotherapy‑free combination of four targeted therapies showed encouraging activity in patients with HR‑positive, HER2‑positive metastatic breast cancer.
The multicenter phase 1/2 ASPIRE trial evaluated anastrozole, palbociclib, trastuzumab, and pertuzumab as a chemotherapy‑free first‑line regimen for HR‑positive, HER2‑positive metastatic breast cancer. In 29 patients, 97 % achieved clinical benefit, median PFS was ~25 months, and median OS had not been reached; 93 % were alive after ~39 months. The safety profile was manageable, with only one treatment‑related discontinuation and no deaths. These findings suggest a potential non‑chemotherapy alternative for a subset of patients.
It offers a less toxic, more convenient first‑line option for metastatic breast cancer patients who may be unsuitable or unwilling to undergo chemotherapy, potentially improving survival and quality of life.
