Mantle cell lymphoma (MCL) is a rare type of non‑Hodgkin B‑cell lymphoma that arises in the mantle zone of lymph nodes. It is characterized by a t(11;14)(q13;q32) translocation leading to cyclin‑D1 overexpression. MCL is usually diagnosed at an advanced stage and is not curable, but a range of therapies – including chemotherapy (e.g., R‑CHOP, Bendamustine‑R), targeted agents (ibrutinib, acalabrutinib, zanubrutinib, Bortezomib), immunotherapy (rituximab), CAR‑T cell therapy, and stem‑cell transplantation – can induce durable remissions. Clinical trials are ongoing to evaluate newer drugs such as pirtobrutinib, Glofitamab, and other combination regimens.

Overexpression of cyclin D1 is a hallmark of mantle cell lymphoma and drives the rapid proliferation of malignant B‑cells, making it a key diagnostic marker and therapeutic target.
