The review summarizes emerging first‑line treatment strategies for TP53‑mutated mantle cell lymphoma (MCL), highlighting data from phase III trials of Bruton tyrosine kinase inhibitors (ibrutinib, acalabrutinib, zanubrutinib) and BCL2 inhibitors (venetoclax), as well as potential MRD‑guided therapy discontinuation and future precision approaches.
TP53‑mutated MCL is associated with aggressive disease and poor prognosis; targeted therapies offer improved survival and may reshape standard care, but optimal duration and long‑term outcomes remain uncertain.
