Purpose: Assess the impact of glucagon‑like peptide‑1 (GLP‑1) receptor agonists on breast cancer risk. Methods: A retrospective cohort study of 111,646 women aged 45‑80 with BMI ≥ 25, identified from electronic health records between 2022 and 2025, using propensity‑matched case‑control design. Results: GLP‑1 agonist exposure was associated with a lower incidence of breast cancer (OR 0.695, 95% CI 0.590‑0.819; P < .0001). Conclusion: GLP‑1 agonists may reduce breast cancer incidence; prospective trials are warranted.
Reduction of breast cancer incidence with GLP‑1 agonists offers a potential preventive strategy that leverages existing diabetes and weight‑management medications, but causal inference requires randomized trials.
