Broad Institute researchers generated 30 new biliary tract cancer cell lines, performed multi‑omic profiling, identified distinct molecular subtypes, and uncovered therapeutic vulnerabilities—including dependence on the EGFR signaling pathway—providing a powerful platform for future drug development.

Broad Institute scientists created 30 new cell lines from biliary tract cancers (BTCs) and performed comprehensive transcriptomic, proteomic, DNA, copy‑number, and CRISPR‑screen analyses. They identified two main subtypes—ductal‑like and bi‑lineage—with distinct transcription factor profiles (HNF1B, GRHL2, KLF5) and uncovered subtype‑specific dependencies, including a broad reliance on the EGFR signaling pathway. These models and discoveries expand the limited BTC resources and pinpoint new therapeutic targets for a disease with poor prognosis.

BTCs are aggressive and lack effective therapies. The new cell line atlas and molecular insights enable targeted drug discovery and provide a platform for preclinical testing, potentially improving treatment options and survival for patients.
