An open‑label study of pemigatinib in cholangiocarcinoma: final results from FIGHT‑202
Pemigatinib demonstrated durable response and prolonged overall survival with manageable adverse events in patients with previously treated, advanced/metastatic cholangiocarcinoma carrying FGFR2 fusions or rearrangements during the extended follow‑up period of the FIGHT‑202 phase II study.
In the FIGHT‑202 phase II trial, 147 patients with advanced/metastatic cholangiocarcinoma harboring FGFR2 fusions or rearrangements received pemigatinib 13.5 mg daily. At median follow‑up of 45.4 months, the objective response rate (ORR) in the FGFR2‑positive cohort was 37.0 %. Median duration of response was 9.1 months, progression‑free survival 7.0 months, and overall survival 17.5 months, with hyperphosphatemia, alopecia, and diarrhea as the most common manageable toxicities.
Cholangiocarcinoma remains a lethal biliary malignancy with limited systemic options. Targeted therapy with pemigatinib exploits FGFR2 fusion/rearrangement biology, providing a clinically meaningful, non‑platinum therapeutic alternative and exemplifying the impact of precision medicine in solid tumours.
