Comparative efficacy of targeted therapy, chemotherapy and their combination for advanced cholangiocarcinoma: a systematic review and network meta-analysis

This systematic review and network meta‑analysis evaluated overall survival (OS) and progression‑free survival (PFS) outcomes in advanced cholangiocarcinoma, comparing chemotherapy (CT), targeted therapy (TT), and combined targeted therapy + chemotherapy (TT+CT) to placebo. All three treatments significantly reduced the hazard ratio (HR) for OS and PFS when compared with placebo; TT+CT yielded the most favourable HR and mean difference (MD) values, and targeted therapy alone uniquely improved PFS.
Demonstrating that these therapeutic strategies can meaningfully prolong survival and delay disease progression informs clinical decision‑making and highlights the added value of combination regimens in advanced cholangiocarcinoma.
