CML is caused by the BCR::ABL1 fusion gene created by the Philadelphia chromosome translocation, which drives uncontrolled granulocytic proliferation.
Chronic myeloid leukemia (CML) is a myeloproliferative disorder driven by the BCR::ABL1 fusion protein resulting from the Philadelphia chromosome translocation. It progresses through chronic, accelerated, and blast phases, and frontline therapy with tyrosine kinase inhibitors (TKIs) such as imatinib, dasatinib, nilotinib, bosutinib, ponatinib, and asciminib has dramatically improved survival.
Early molecular diagnosis and targeted TKI therapy transform CML from a fatal disease to a chronic, treatable condition with near‑normal life expectancy for most patients.
