CD151 Promotes Cancer Progression in Triple‑Negative Breast Cancer by Inducing EMT through the MAPK Signaling Pathway
CD151 promotes migration, invasion and epithelial‑mesenchymal transition in triple‑negative breast cancer (TNBC) by binding to integrin α3β1 and activating the MAPK signaling pathway, thereby acting as a pro‑tumorigenic factor.
This study demonstrates that CD151 is highly expressed in TNBC tissues and cell lines and that its over‑expression enhances the migratory and invasive abilities of TNBC cells through the induction of epithelial‑mesenchymal transition (EMT). Co‑immunoprecipitation experiments reveal that CD151 binds to integrin α3β1, and Western blot analysis shows that this complex activates the MAPK signaling pathway (ERK1/2 and JNK), which in turn drives EMT. Knockdown of CD151 or ITGA3/ITGB1 reduces migration and invasion in vitro. These findings suggest that CD151 functions as a novel driver of TNBC progression and could serve as a potential therapeutic target.
Targeting CD151 may impede EMT and subsequent metastasis in TNBC, a subtype with limited treatment options and poor prognosis, thereby improving therapeutic outcomes.
