Pancreatic adenocarcinoma remains one of the deadliest malignancies, with a low 5‑year survival rate. This review summarizes the history and current landscape of phase III clinical trials in advanced pancreatic cancer, highlighting the modest success rate of translating trial results into standard care and the growing need for biomarker‑guided, stratified studies to improve trial efficiency.
Optimizing clinical trial design and incorporating predictive biomarkers may accelerate the development of effective therapies for pancreatic cancer, potentially improving patient survival and reducing the burden on limited trial resources.
