ASTRIS, a large real-world study to evaluate the efficacy of osimertinib in epidermal growth factor receptor T790M mutation-positive non-small cell lung cancer patients: Clinical characteristics and genotyping methods in a Spanish cohort

Osimertinib has proven efficacy in EGFR T790M mutation‑positive non‑small cell lung cancer (NSCLC) patients; however, its benefits have not been evaluated in a real‑world setting. ASTRIS is expected to confirm these benefits in routine clinical practice.

Purpose: Osimertinib has proven efficacy in EGFR T790M mutation-positive non‑small cell lung cancer (NSCLC) patients; however, its benefits have not been evaluated in a real-world setting. Methods: ASTRIS is a single‑arm, open‑label, multinational study evaluating the efficacy and safety of osimertinib for treatment of EGFR T790M mutation‑positive NSCLC. Results: The Spanish cohort included 131 patients (68.7% women, 97.7% Caucasian, mean age 64.8 years). Forty patients (28.1%) remained on therapy at cutoff. Tissue samples (55.0%) and primary tumor sites (61.1%) were most common for T790M testing; local laboratories performed 60.3% of analyses; Roche Cobas EGFR assay 43.5%. Conclusions: ASTRIS is expected to confirm the benefits of osimertinib in a real‑world setting and provide additional information on best practices for detecting EGFR T790M mutations.

Real‑world evidence will confirm osimertinib’s efficacy outside the controlled environment of clinical trials and guide optimal detection of EGFR T790M in routine practice.
