The survival rates for osteosarcoma have remained essentially unchanged over the past three decades despite attempts to improve outcomes via dose intensification and modification based on response. This review discusses recent findings from contemporary clinical trials, advances in understanding osteosarcoma biology and genomic complexity, and potential opportunities using targeted and immune‑mediated therapies.
Osteosarcoma is the most common primary bone malignancy in children and adolescents. Current therapies based on cytotoxic agents have plateaued, and novel therapeutic strategies informed by genomic and immunologic insights are urgently needed to improve survival, especially for patients with metastatic or refractory disease.
