KRAS G12C‒specific inhibitors (adagrasib, sotorasib) with EGFR inhibitors are approved for metastatic colorectal cancer with KRAS G12C mutations after prior chemotherapy.
The KRAS mutation is present in about 40‑45% of colorectal cancers. Testing for KRAS (and NRAS) is recommended in metastatic colorectal cancer to guide the use of EGFR‑targeted therapies (cetuximab, panitumumab). KRAS G12C‑specific inhibitors (adagrasib, sotorasib) in combination with EGFR inhibitors are FDA‑approved for KRAS G12C‑mutant colorectal cancer after prior chemotherapy. Ongoing trials explore agents for other KRAS mutations and novel combination regimens.
KRAS status determines eligibility for EGFR blockers and KRAS‑specific therapies, influencing survival and treatment decisions in metastatic colorectal cancer.
