Head and Neck Cancer Clinical Benefit and Safety of Reduced Elective Dose in Definitive Radiotherapy for Head and Neck Squamous Cell Carcinoma: The UPGRADE-RT Multicenter Randomized Controlled Trial

Rákkutatás · Head and neck squamous cell carcinoma

In this multicenter randomized trial, the clinical benefit and safety of definitive RT with reduced versus standard elective dose were compared.

The phase 3 KEYNOTE‑689 trial evaluated the addition of pembrolizumab to standard therapy (surgery followed by radiation or radiation plus chemotherapy) in patients with locally advanced, previously untreated stage 3–4 HNSCC whose tumors expressed PD‑L1. At 36 months, event‑free survival was 60 % in the pembrolizumab arm versus 46 % in the control arm, representing a 27 % relative reduction in recurrence risk.
