Prostate cancer: Three drug trials to watch
Olaparib (Lynparza) and rucaparib (Rubraca) are FDA‑approved for the treatment of metastatic, castration‑resistant prostate cancer in men with BRCA1, BRCA2, or other DNA‑repair gene mutations.
The article reports on a Phase 2 pilot study evaluating the feasibility and potential benefit of administering the PARP inhibitor niraparib (ZEJULA) as a neoadjuvant therapy for men with high‑risk, aggressive prostate cancer. Eleven patients with specific DNA repair gene mutations received 200 mg daily for 90 days prior to surgery. Although the drug did not produce significant tumor shrinkage, the study highlighted the utility of genetic profiling and circulating tumor DNA monitoring to better understand disease biology and resistance mechanisms. The findings suggest a role for targeted neoadjuvant approaches and underscore the need for larger, randomized trials.
These approvals provide new targeted therapeutic options for men with metastatic prostate cancer who have DNA‐repair deficiencies, potentially improving progression‑free and overall survival and expanding personalized medicine in this setting.
