Lifileucel Confers Durable Responses in Pretreated Advanced NSCLC

Lifileucel monotherapy achieved an objective response rate of 25.6% and durable responses in previously treated advanced nonsquamous NSCLC patients.

In a registrational phase‑2 trial (IOV‑LUN‑202, NCT04614103), 39 patients with metastatic or unresectable nonsquamous NSCLC who had progressed on prior chemotherapy and immune‑checkpoint inhibitors received lifileucel (Amtagvi) monotherapy following non‑myeloablative lymphodepletion and interleukin‑2 support. The objective response rate (RECIST v1.1) was 25.6% (10/39), including 2 complete and 7 partial responses. The disease‑control rate was 71.8%. With a median follow‑up of 25.4 months, the median duration of response was not reached, demonstrating durable disease control. The safety profile was consistent with lymphodepletion and IL‑2, with no new safety signals.

The study indicates a meaningful clinical benefit in a historically hard‑to‑treat population of NSCLC patients who have failed standard therapies, offering a one‑time cellular therapy that yields durable responses and could expand therapeutic options.
