Ewing Sarcoma Treatment
Ribociclib combined with everolimus elicits tumor responses in patients with advanced dedifferentiated liposarcoma.
A phase 2 trial (SAR-096, NCT03114527) evaluated ribociclib plus everolimus in patients with advanced dedifferentiated liposarcoma (DDLPS). The combination produced disease stabilization and two confirmed partial responses among 21 enrolled patients, with a tolerable safety profile. Biomarker analyses are planned to identify predictive markers for response.
This finding provides evidence that dual CDK4 and mTOR inhibition can produce objective responses in a sarcoma subtype that has shown limited activity with CDK4 inhibitors alone, offering a potential new therapeutic option for DDLPS patients.
