A combination of talazoparib and enzalutamide reduced the risk of disease progression or death by 52% compared with enzalutamide alone in patients with metastatic castration‑sensitive prostate cancer bearing BRCA1 or BRCA2 mutations.
In patients with metastatic castration‑sensitive prostate cancer harboring BRCA1/2 mutations, combining talazoparib with enzalutamide produced a 52% relative reduction in risk of disease progression or death versus enzalutamide alone. After ≥3 years, radiographic progression‑free survival was 77% with the combination versus 56% with monotherapy, without significant decline in patient‑reported quality of life.
Delaying progression to castration‑resistant disease remains a key challenge in early metastatic prostate cancer, especially for patients with BRCA1/2 mutations associated with poorer outcomes.
