Phase II Trial of Chemotherapy, Cetuximab, and Erlotinib in Patients With Metastatic or Recurrent Squamous Cell Carcinoma of the Head and Neck
Adding erlotinib to cetuximab and chemotherapy improves objective response rates, progression‑free survival, and overall survival in patients with metastatic or recurrent squamous cell carcinoma of the head and neck.
In a 24‑patient, phase II study of metastatic or recurrent HNSCC, patients received carboplatin and paclitaxel with cetuximab and, from cycle 2 onward, erlotinib. The addition of erlotinib increased objective response rate from 33.3% to 58.3%, median progression‑free survival was 6.2 months, and median overall survival was 10.6 months.
HNSCC has limited treatment options and poor survival rates. Enhancing response to therapy could improve survival and quality of life for these patients.
