Sigvotatug vedotin, an investigational IB6-directed antibody‑drug conjugate, did not significantly improve overall survival versus docetaxel among patients with previously treated locally advanced, unresectable, or metastatic nonsquamous non‑small cell lung cancer (NSCLC).

Phase 3 SigVie‑002 evaluated sigvotatug vedotin versus docetaxel in previously treated, locally advanced or metastatic nonsquamous NSCLC. The primary endpoint overall survival was not met across the overall population, although second‑line patients showed a stronger trend for OS and progression‑free survival. Exploratory analyses found no clear link between IB6 expression and response; safety was comparable to prior reports.

Although sigvotatug vedotin failed to demonstrate a survival benefit overall, its activity in second‑line treatment and potential synergy with immunotherapy warrant continued investigation in earlier lines, where docetaxel remains a meaningful comparator.
