Twenty Years of Advancing Discoveries and Treatment of Mantle Cell Lymphoma
Mantle cell lymphoma is a heterogeneous disease whose treatment response is influenced by genetic mutations, epigenetic alterations, and the tumor microenvironment, and which is experiencing new therapeutic options including targeted kinase inhibitors, bispecific antibodies, and CDK4/6 + BTK combination strategies.
Mantle cell lymphoma (MCL) is an aggressive B‑cell lymphoma with a characteristic t(11;14) translocation leading to cyclin D1 overexpression. It is highly heterogeneous at the genetic, epigenetic, and microenvironmental levels, resulting in variable clinical outcomes and limited standard of care. Recent advances include the identification of novel therapeutic targets, the development of bispecific antibodies, and combination strategies with CDK4/6 inhibition and BTK inhibitors. Ongoing clinical trials aim to validate biomarkers and improve treatment efficacy.
Understanding the molecular drivers of MCL heterogeneity and therapeutic resistance is essential for tailoring individualized treatments, improving survival rates, and ultimately moving toward a cure for this aggressive lymphoma.
