New strategies in ovarian cancer treatment

Ovarian cancer cells survive platinum‐ and taxane‐based chemotherapy by reprogramming their metabolism, and matching metabolic inhibitors to the dominant resistance state can resensitize these tumours to treatment.

A comprehensive review highlights that ovarian cancer chemoresistance is driven by metabolic reprogramming of glycolysis, mitochondrial respiration, lipid metabolism, ferroptosis, and cuproptosis. The authors argue that each tumour may adopt distinct dynamic metabolic states that enable survival against platinum–taxane therapy and that targeting these specific metabolic vulnerabilities in combination with chemotherapy could overcome resistance.

If resistance is rooted in tumour metabolism, then personalised metabolic profiling could guide rational drug combinations, potentially improving outcomes for patients with platinum‑resistant ovarian cancer.
