Amivantamab Monotherapy in Chemorefractory RAS / BRAF Wild‑Type Metastatic Colorectal Cancer: Results From OrigAMI‑1, an Open‑Label, Phase Ib/II Study
Amivantamab monotherapy demonstrates promising, durable antitumor activity in chemorefractory metastatic colorectal cancer irrespective of prior anti‑EGFR therapy and primary tumour location.
Amivantamab, an EGFR‑MET bispecific antibody, was evaluated in the phase Ib/II OrigAMI‑1 study for chemorefractory metastatic colorectal cancer (mCRC) with RAS/BRAF wild‑type status. At a median follow‑up of 11.9 months, objective response rates ranged from 19% to 29% in left‑sided cohorts and 22% in the right‑sided cohort, with durable responses (median duration 9.0–9.8 months) and a manageable safety profile. These results support further investigation of amivantamab in combination with chemotherapy and in earlier lines of treatment.
Metastatic colorectal cancer with RAS/BRAF wild‑type lacks actionable targets beyond anti‑EGFR agents, which have limited efficacy in late‑line settings. Demonstrating activity of a bispecific EGFR‑MET inhibitor offers a novel therapeutic avenue and may improve outcomes for patients with chemorefractory disease.
