A Major Milestone in Glioblastoma Research
Preliminary clinical trial results from Mass General Cancer Center show ‘dramatic and rapid’ regression of glioblastoma after next‑generation CAR T therapy.
Mass General Cancer Center’s INCIPIENT phase 1 trial evaluated a novel CAR‑TEAM‑E T‑cell therapy (CARv3‑TEAM‑E) combining CAR T‑cells with bispecific T‑cell‑engaging antibodies against EGFR. In three recurrent glioblastoma patients, a single intracerebral infusion induced dramatic tumor shrinkage, with one patient achieving near‑complete regression. Toxicity was manageable, though all patients experienced fevers and altered mental status. Despite initial responses, all patients developed tumor progression within months, likely due to limited persistence of the engineered cells.
These early results demonstrate that a combined CAR T + bispecific antibody approach can overcome the heterogeneity of glioblastoma and achieve rapid, profound tumor regression, offering a new therapeutic avenue for this lethal brain cancer.
