SWOG study shows strong long-term survival rates for patients with GIST
The PDGFRα antibody olaratumab showed activity in cell line xenograft models and an initial phase II trial, but failed to improve survival in a definitive phase III STS trial.
This phase 2 single‑arm study evaluated cabozantinib in 50 metastatic GIST patients after failure of imatinib and sunitinib. At 12 weeks, 24 of 41 evaluable patients were progression‑free, exceeding the predefined threshold. Median progression‑free survival was 6 months, disease‑control rate 80 %, and no new safety signals were observed.
The findings indicate cabozantinib could become an alternative therapy for GIST patients who have progressed on standard TKIs, potentially overcoming MET‑mediated resistance and improving clinical outcomes.
