Rinatabart sesutecan (Rina‑S) Shows Promising Antitumor Activity in Advanced Ovarian Cancer

Rinatabart sesutecan (Rina‑S) produced encouraging clinical activity, with overall response rates of 22.7% at 100 mg/m² and 55.6% at 120 mg/m², and disease‑control rates of 86–89% in patients with platinum‑resistant ovarian cancer.

Phase 1/2 RAINFOL‑01 dose‑expansion part B evaluated rinatabart sesutecan, a folate receptor α‑directed antibody‑drug conjugate, in 42 patients with platinum‑resistant ovarian cancer. Median follow‑up was 46–48 months. Confirmed overall response rates were 22.7% at 100 mg/m² and 55.6% at 120 mg/m². Disease‑control rates exceeded 86% in both cohorts, with median duration of response not reached. The drug was well tolerated, with manageable grade 3/4 toxicities, and no new safety signals were reported.

The results suggest that targeting folate receptor α with an antibody‑drug conjugate can produce deep, durable responses in a population with limited treatment options. If confirmed in larger trials, this therapy could offer a new, effective strategy for platinum‑resistant ovarian cancer.
