ECHO trial 50-month follow‑up results: ABR vs PBR in TN MCL

Median progression‑free survival was significantly longer with acalabrutinib‑based therapy (ABR) compared with placebo‑based therapy (PBR) (72.5 vs 47.8 months).

The ECHO trial (NCT02972840) was a phase III randomized controlled study evaluating acalabrutinib plus bendamustine and rituximab (ABR) versus placebo plus bendamustine and rituximab (PBR) as first‑line therapy in 598 older patients (≥65 years) with treatment‑naïve mantle cell lymphoma. Updated 50‑month follow‑up demonstrated superior median progression‑free survival with ABR versus PBR (72.5 vs 47.8 months; HR 0.68; 95% CI 0.53–0.87; p = 0.0022).

First‑line treatment with ABR significantly delayed the need for subsequent therapies and extended PFS versus PBR in older patients with TN MCL, with no new safety signals, supporting its use as a first‑line therapy in this patient population.
