Ivonescimab Plus Chemotherapy Global Phase III HARMONi Primary Analysis Results Published in The Lancet Oncology

Cancer research · NSCLC · EGFR

In HARMONi, ivonescimab plus chemotherapy demonstrated a statistically significant and clinically meaningful improvement in progression‑free survival (PFS) compared to placebo plus chemotherapy in patients with EGFR‑mutated non‑small cell lung cancer (NSCLC) after progression on third‑generation EGFR tyrosine kinase inhibitors (TKIs).

In the global Phase III HARMONi trial, ivonescimab in combination with platinum‑doublet chemotherapy significantly improved progression‑free survival (median 6.8 months vs 4.4 months) versus placebo plus chemotherapy in patients with EGFR‑mutated locally advanced or metastatic non‑small cell lung cancer (NSCLC) who progressed after a third‑generation EGFR TKI. The benefit was consistent across pre‑planned subgroups and represents a clinically meaningful advance for a population with limited post‑TKI options.

Patients with EGFR‑mutated NSCLC who progress on third‑generation EGFR TKIs have few effective treatment options. The demonstrated PFS benefit of ivonescimab plus chemotherapy offers a new therapeutic strategy that targets both PD‑1 and VEGF, potentially improving outcomes where current therapies have limited benefit.
