The addition of daratumumab to the KRd regimen is a new standard of care for newly diagnosed multiple myeloma patients.
A new four‑drug combination (daratumumab + carfilzomib + lenalidomide + dexamethasone) has shown superior efficacy and safety compared with the three‑drug KRd regimen in the ADVANCE randomized trial of newly diagnosed multiple myeloma patients, supporting the addition of daratumumab as a new standard of care.
In a randomized, multi‑center trial involving 306 patients, daratumumab‑based therapy (DKRd) achieved a higher MRD‑negative rate (59% vs 36%) and improved progression‑free survival (86% vs 79%) without increased toxicity, indicating a safer and more effective treatment option for patients who would otherwise receive only KRd.
