Olaparib Maintenance Therapy Delays Ovarian Cancer Progression
Olaparib, a PARP inhibitor, significantly delays progression of advanced ovarian cancer in women with BRCA mutations when used as maintenance therapy after initial chemotherapy.
In the SOLO‑1 phase 3 randomized trial, 386 women with newly diagnosed advanced ovarian cancer carrying BRCA1/2 mutations received olaparib or placebo after completing standard platinum‑based chemotherapy. Median follow‑up was 41 months with a median progression‑free survival of over 4 years in the olaparib group versus 14 months in the placebo group, yielding a 70 % reduction in the risk of progression or death. The drug was well tolerated, with only 12 % discontinuations due to adverse events.
These results establish olaparib as a highly effective maintenance therapy that extends disease control in a defined genetic subgroup, offering clinicians a new standard-of-care option for patients with BRCA-mutated ovarian cancer and potentially improving long‑term outcomes.
