Retifanlimab combined with carboplatin–paclitaxel improves overall survival versus placebo in patients with advanced squamous cell anal cancer

In the phase III POD1UM-303/InterAACT-2 study, first‑line retifanlimab plus carboplatin–paclitaxel (CP) achieved a 25% relative reduction in death risk compared with placebo + CP in 308 patients with advanced squamous cell anal cancer (SCAC). Median overall survival (OS) was 32.8 months versus 22.2 months (median follow‑up 26.3 vs 20.6 months). The benefit was consistent across most predefined subgroups.

This is the first phase‑III randomized trial demonstrating a clinically meaningful OS advantage with a PD‑1 inhibitor plus chemotherapy as standard care for advanced SCAC, a rare disease with historically poor prognosis.
