Evidence of being cured for nasopharyngeal carcinoma: results of a multicenter patient-based study in China

Statistical cure is potentially achievable among patients with NPC undergoing contemporary treatment modalities.

This large‑scale retrospective multicenter cohort study included 6,315 non‑metastatic nasopharyngeal carcinoma (NPC) patients from four tertiary cancer centres in China (2007–2020). Patients received intensity‑modulated radiotherapy (IMRT) with optional platinum‑based chemotherapy. Using mixture and non‑mixture cure models that incorporate background population mortality, the probability of achieving a life expectancy equivalent to the general population was 78.1 % when death alone was the uncured event and 72.4 % when progression (death, recurrence, or metastasis) was considered the uncured event. The time required for 95 % of patients to reach statistical cure was 7.1 years for the death model and 4.7 years for the progression model. Cure probabilities were higher in younger patients, those with better Eastern Cooperative Oncology Group performance, lower TNM stage, lower Epstein‑Barr virus DNA copies and lower lactate dehydrogenase levels. The study demonstrates that contemporary treatment can achieve statistical cure in a large proportion of NPC patients.

This comprehensive evidence confirms that nasopharyngeal carcinoma can be considered curable with modern therapeutic approaches. The findings support refined patient counseling, may inform the design of future clinical trials, and highlight the importance of risk‑stratified management strategies.
