Idecabtagene Vicleucel in Relapsed and Refractory Multiple Myeloma
Idecabtagene vicleucel induces a 73% overall response rate in heavily pre‑treated patients with relapsed and refractory multiple myeloma.
In this phase 2 KarMMa study, idecabtagene vicleucel (ide‑cel, bb2121) was evaluated in 128 patients with relapsed and refractory multiple myeloma after ≥3 prior regimens. The overall response rate was 73% (94/128) and the CR/CRi rate was 33% (42/128). MRD‑negative status (<10‑5) was achieved in 26% of treated patients. Median progression‑free survival was 8.8 months. Common toxicities included neutropenia (91%), anemia (70%), thrombocytopenia (63%), cytokine release syndrome (84% – 5% grade ≥ 3) and neurotoxicity (18% – 3% grade 3).
Multiple myeloma that is refractory to standard therapies has limited treatment options; ide‑cel represents a novel CAR‑T therapy that has demonstrated meaningful clinical responses in patients with very few options, potentially improving survival and quality of life.
