Deciphering the potential pathogeny of rare tracheal adenoid cystic carcinoma by single‑cell RNA‑sequencing

Developing drugs that target MIF‑CD74 signaling pathways may lead to breakthroughs in the treatment of TACC

This study performs single‑cell RNA‑sequencing on tumor, adjacent tissue, and blood from a patient with tracheal adenoid cystic carcinoma (TACC). Analysis identified distinct epithelial, immune, and stromal clusters, revealing that ciliated epithelial cells with stem‑cell markers likely represent the tumor’s origin. Immunophenotyping and multiplex immunohistochemistry confirmed aberrant expression of CD68, CD74, CD44, CXCR4, Vimentin, and PANCK, and highlighted interactions between mesothelial‑like malignant cells and macrophages via MIF‑associated pathways. These findings suggest that targeting MIF‑CD74 signaling could form the basis for novel therapeutic strategies in TACC.
