Decoding T-Cell Lymphoma: How Molecular Biology Is Driving a New Era of Targeted Treatment

Molecular biology insights are guiding the development of targeted therapies that improve outcomes in peripheral T‑cell lymphoma (PTCL).

Peripheral T‑cell lymphoma (PTCL) represents a heterogeneous group of rare, aggressive lymphomas with poor prognosis. Recent research at Memorial Sloan‑Kettering has leveraged molecular profiling to identify key pathways—PI3K, JAK/STAT, EZH2 dysregulation—and develop targeted agents such as the anti‑CD94 antibody DR‑01, the PI3K inhibitors duvelisib, and the EZH2 inhibitor valemetostat. Early phase trials, including a phase 2 study (NCT05475925) and the duvelisib PRIMO trial, demonstrate promising overall response rates and pave the way for combinatorial approaches to address toxicity and resistance.

Improved targeted treatment strategies based on molecular biology have the potential to increase response rates, extend survival, and reduce the severe toxicities associated with conventional cytotoxic chemotherapy for patients with PTCL.
