Cytotoxic effects of targeted agent alone or with chemotherapy in the treatment of adenoid cystic carcinoma: a preclinical study
Data obtained demonstrated that both vorinostat and olaparib significantly increased the standard chemotherapy cytotoxic effects, suggesting new interesting therapeutic options for ACC.
Adenoid cystic carcinoma (ACC) is a rare malignancy with high relapse rates and poor long‑term prognosis. Preclinical evaluation of cytotoxic effects of standard chemotherapeutic agents (cisplatin, doxorubicin) alone and in combination with five targeted therapy drugs (vorinostat, olaparib, palbociclib, everolimus, lenvatinib) was performed in vitro on an h‑TERT immortalised ACC cell line and in vivo using a zebrafish xenograft model. The combination of vorinostat or olaparib with cisplatin or doxorubicin produced synergistic cytotoxicity, markedly enhancing tumour growth inhibition in the zebrafish model. These findings suggest new therapeutic options for relapsed or metastatic ACC.
This study identifies vorinostat and olaparib as promising agents capable of synergistically enhancing the efficacy of conventional chemotherapeutic drugs in adenoid cystic carcinoma, potentially improving treatment outcomes for this otherwise hard‑to‑treat malignancy.
