Ruxolitinib is Under Investigation as a JAK Inhibition Strategy for Multiple Myeloma

The observed clinical benefit of ruxolitinib concomitant with methylprednisolone and lenalidomide in relapsed/refractory multiple myeloma was an overall response rate (ORR) of 31% for the initial 2‑drug combination and 30% once lenalidomide was added; median progression‑free survival (PFS) remained 3.5 months regardless of additional therapy.

These data support evaluation of JAK inhibition as a therapeutic avenue in refractory myeloma, a disease with limited options after standard salvage regimens.
