This study identified a macrophage subtype expressing SPP1 and TREM2 that promotes immune suppression and metastasis in advanced prostate cancer, particularly in bone metastases. Blocking SPP1 with an antibody in mouse models increased tumor susceptibility to immunotherapy, underscoring the therapeutic potential of targeting this macrophage population.
Targeting SPP1/TREM2‑positive macrophages offers a strategy to overcome the resistance of prostate cancer to immunotherapy, potentially improving outcomes for patients with metastatic disease.
