Chronic myelogenous leukemia (CML) is a clonal myeloproliferative disorder resulting from the neoplastic transformation of the primitive hematopoietic stem cell.
Chronic myelogenous leukemia (CML) is a clonal myeloproliferative disorder characterized by the Philadelphia chromosome (t(9;22)(q34;q11)) and the BCR‑ABL fusion protein with constitutive tyrosine‑kinase activity. The disease progresses through chronic, accelerated and blastic phases, and imatinib (Gleevec) has become the standard of care, significantly improving survival and reducing mortality.
CML is the world’s most common form of acute leukemia in adults with a rising incidence. Late-stage disease can transform into a blast crisis, where long‑term survival is often <6 months. Early identification of the Philadelphia chromosome and the introduction of BCR‑ABL‑specific inhibitors such as imatinib have dramatically changed the natural history of CML, allowing most patients to reach a clinically meaningful survival of 10–15 years when adequately treated.
