This study investigates the association between thyroid cancer and weight‑loss medications, focusing on the FDA Adverse Event Reporting System (FAERS) data from 2004 to Q1 2024. A disproportionality analysis identified significant positive associations between GLP‑1 receptor agonists (semaglutide, dulaglutide, liraglutide, and tirzepatide) and reported thyroid cancer cases, with reporting odds ratios ranging from 2.09 to 15.59. No significant signals were seen for other class members such as empagliflozin, dapagliflozin, or metformin, which showed a weak inverse association. The findings underscore the need for ongoing safety monitoring of GLP‑1 receptor agonists when used for obesity management.
Thyroid cancer can be life‑threatening and currently‑approved weight‑loss therapeutics such as semaglutide and tirzepatide are widely prescribed. Detecting a potential signal of increased risk early enables clinicians to adjust treatment plans, informs regulatory updates, and promotes patient safety in a rapidly expanding therapeutic area.
