Effect of Daratumumab on Patients with High-Risk Cytogenetic Multiple Myeloma: A Systematic Review and Meta‑Analysis of Randomized Controlled Trials

In patients with high‑risk cytogenetic multiple myeloma, adding daratumumab to standard therapy improves progression‑free survival compared with standard therapy alone.

A meta‑analysis of 15 randomized controlled trials demonstrated that daratumumab significantly improves progression‑free survival (PFS) in patients with high‑risk cytogenetic multiple myeloma (HR = 0.58, 95% CI 0.49‑0.69, P < 0.001).

Improved PFS may translate into longer periods of disease control and potentially better overall survival for a population known to have a poor prognosis, informing clinicians and guideline developers about the benefits of incorporating daratumumab into frontline and relapsed regimens.
