Lamin subtype expression in Merkel‑cell carcinoma: its relation to polyomavirus infection and tumor metastasis

Cancer research · NSCLC

Primary MCC tumors express variable, but mostly low levels of A‑type lamins, while the B‑type lamins are overall highly expressed.

Merkel‑cell carcinoma (MCC) is a rare, highly aggressive skin cancer with a strong metastatic potential. In several tumors, A‑ and B‑type lamins have been linked to invasion and metastasis, yet MCC has not been studied in this context. We examined lamin expression in 38 FFPE tissue samples from 28 MCC patients (primary tumours and metastases) using immunohistochemistry. Primary MCCs show low A‑type but high B‑type lamin expression, regardless of MCPyV‑status. Metastatic lesions occasionally exhibit increased A‑type lamin expression, but paired primary–metastatic comparisons reveal no consistent change. Normal Merkel cells express both lamin types. These data suggest a flexible A‑type lamin expression during MCC carcinogenesis and metastasis, underscoring the aggressiveness of MCC and contributing to the debate on its cellular origin.

The lamina composition may influence nuclear mechanics, potentially affecting MCC invasion and metastatic capacity.
