Mouse and human urothelial cancer organoids: A tool for bladder cancer research
Established a living biobank of bladder cancer organoids from over 50 patients that mirrors tumor pathogenesis and can be used for drug screening.
Bladder cancer poses a major clinical challenge, especially in advanced stages. Here we present a culture system for bladder cancer based on organoid technology, establishing a long‑term culture system for both normal mouse urothelium and human bladder cancer cells. We created a living biobank of organoids derived from 53 patient samples, preserved in 3D structures that recapitulate the tumor histology and include basal and luminal subtypes. Common driver mutations such as TP53 and FGFR3 were identified in the biobank. Limited drug testing on these organoids demonstrates their potential as a platform for screening and personalized medicine.
The creation of a diverse and genetically representative biobank provides an essential in vitro model to study bladder tumor biology, identify therapeutic vulnerabilities, and accelerate the development of targeted treatments for melanoma and urothelial cancers.
