Kirsten rat sarcoma viral oncogene (KRAS) G12C inhibitors have demonstrated clinical efficacy against KRAS G12C‑mutant non‑small cell lung cancer (NSCLC); however, intrinsic and acquired resistance limit their therapeutic potential. Combination strategies targeting EGFR-mediated pathways may overcome this resistance.
Resistance to KRAS G12C inhibitors reduces treatment effectiveness in NSCLC patients harboring the KRAS G12C mutation. Combining EGFR inhibition with KRAS targeting may restore drug sensitivity and improve clinical outcomes.
